Hi, the additional base plate/pickup mounting plate for adapting the B5 to a Tele has extra screw holes for mounting the plate right where you don't want them if you were going to machine out a humbucker sized hole. You could possibly hide these by covering them with a flat metal humbucker surround ring (which may fit depending on the width dimension of the plate and the ring) on top of the plate which would also mount the humbucker or you could simply dispense with the plate altogether and mount the humbucker like any other - the bridge is a 'Jazzmaster' style unit by the looks and I would upgrade it if possible (this bridge sucks big time and is impossible to keep in tune with a trem) to a Schaller roller or something similar with the correct spring spread and mount the bridge like a tunomatic.
The modern Gretch rock guitars (Elliot Easten) use a similar combination to keep in tune and get some attack going.

Alternatively, if you are driven by the looks and the romance of crappy engineering from the 50's and wish to maintain the base plate unit I would suggest locking up the original bridge as per the Jazzmaster mod pages which fixes the bridge locating pins by sleeves or muti wrappings of shielding foil (it works) - and shop around for a single coil sized humbucker such as the Dimarzio Fast track/Chopper (which I rate) or the Duncan range of 'littles' ...they work and punch hard for their weight. The Dimarzios are my pick for a Ash/Alder bodied Fender style instrument because I know they work well tonally and they split (coil split) very nicely.
